abstract

objectives: in present research we evaluate the expression of this critical enzyme in a eukaryotic system for future use in cheese industry. materials and methods: we have cloned bovine prochymosin gene in methylotrophic yeast, p. pastoris, using ppic9k as an expression vector. the recombinant plasmid was transformed into the host by electroporation, and it was expressed in optimum conditions (temperature 29oc, 200 rpm, 2% methanol for induction, and 5 days of incubation). transcription and expression of the recombinant prochymosin was evaluated by the reverse transcription polymerase chain reaction (rt-pcr), sodium dodecyl sulfate polyacrylamide gel electrophoresis (sds-page) analysis as well as western blotting and enzyme-linked immunosorbent assay (elisa). results: in optimum conditions, only a low level of this heterologous protein was detected using elisa method and subsequently confirmed by rt-pcr. conclusions: since it has been reported that p. pastoris is an appropriate host for the expression of recombinant proteins, a low level of expression of prochymosin in this host should be explored in our future research.

Cloning and heterologous expression of Laccase in pichia pastoris and determination some of biochemical properties

Laccase (EC 1.10.3.2) are multi-copper oxidase which catalyze the oxidation aromatic and non- aromatic compounds with electron reduction of molecular oxygen to water. Nucleotide sequence of laccase (accession number : ) was optimized according codon preference of Pichia pastoris. Gene was synthesized and cloned into pPICZalpha A. laccase under control of AOX1 promoter was transformed to P.pasto...

Heterologous expression of the antimyotoxic protein DM64 in Pichia pastoris

Snakebite envenomation is a neglected condition that constitutes a public health problem in tropical and subtropical countries, including Brazil. Interestingly, some animals are resistant to snake envenomation due to the presence of inhibitory glycoproteins in their serum that target toxic venom components. DM64 is an acidic glycoprotein isolated from Didelphis aurita (opossum) serum that has b...

Heterologous protein expression in the methylotrophic yeast Pichia pastoris.

During the past 15 years, the methylotrophic yeast Pichia pastoris has developed into a highly successful system for the production of a variety of heterologous proteins. The increasing popularity of this particular expression system can be attributed to several factors, most importantly: (1) the simplicity of techniques needed for the molecular genetic manipulation of P. pastoris and their sim...

Heterologous protein production using the Pichia pastoris expression system.

The Pichia pastoris expression system is being used successfully for the production of various recombinant heterologous proteins. Recent developments with respect to the Pichia expression system have had an impact on not only the expression levels that can be achieved, but also the bioactivity of various heterologous proteins. We review here some of these recent developments, as well as strateg...

P-197: Expression of Bovine Follicle Stimulating Hormone Subunits in Pichia Pastoris

Background: Bovine follicle stimulating hormone (bFSH) belongs to the family of glycoprotein hormones that are released from the pituitary gland or the placenta. This hormone is a heterodimer consisting of common α- and specific β-subunit. Bovine FSH is used for reproductive technology such as superovulation in farm animals and polycystic ovary syndrome treatment. Also this hormone is produced ...
